研究概述

Diabetes Mellitus (DM) is one of the aging associated diseases and has been intensely focused recently. My research interest is on regenerative medicine - mainly to investigate the advert effect of diabetic physiology on different cells and how to re-grow missing tissues resulted from diabetic insults for replacement therapy in cellular and molecular basis. To this end, the projects in my laboratory include: (1) To explore molecular cues underlying DM mediated tissue impairment in normal, neoplastic and stem cells and (2) To elucidate signaling modulators of candidate pathways reverse pathological conditions. In clinic, under the collaboration with medical doctors, the exploration of disease predictors would be my desire in order to prevent clinical disorders.

Several specific questions to be addressed in my laboratory: 1. The intrinsic and extrinsic impacts of glucose/lipid metabolic regulation on head and neck carcinogenesis * Mostly from epidemiological studies, diabetes potentially regulates carcinogenesis in various cancers. The conclusive association between diabetic/hyperglycemic condition and oral head and neck cancer onset/progression, however, is still unclear. In addition, whether the reversal of canerous metabolism modulate head and neck cancer malignancy is also one of the candidate projects in my laboratory. We therefore aim to identify the underlying mechanisms related to this association, both in vitro and in vivo. The outcome would be of great benefit to provide essential information understanding how metabolic imbalance regulates malignancy, or vice versa, and to establish a useful platform for targeting cancer metabolism for development of therapeutic anti-cancer agents. 2. The intrinsic and extrinsic cellular and molecular cues for (re-)generate functional liver and pancreatic cells from isolated stem cells or endogenous pluripotent stem/progenitor cells ** Maintenance of normal physiological function could be achieved by proper “cross-talk” between cells and surrounding environment. The examination of dynamic molecular players during tissue loss or regeneration is important to uncover essential factors in maintaining normal physiology. In the aspect of translational medicine, to understand the local and systemic molecular regulations could be useful to provide unlimited sources for clinical practice such as organ transplantation. 3. The effect of hyperglycemic/diabetic physiology on tissue homeostasis *** Adult stem cells reside in different tissues and are believed to play an important role in restoring impaired tissues when needed. We aim to understand how diabetic pathology affects cell properties of adult stem cells.
