電位控制鉀離子通道(Voltage-gated potassium channels)參與神經系統的許多生理功能。這些電位控制鉀離子通道具多樣性,且在細胞和次細胞層次有差異性表現,因而能在特定神經細胞產生特別的電性質。除了在腦部表現量豐富,目前對ether-à-go-go(EAG)鉀離子通道在神經細胞的生理功能並不清楚。為了更進一步了解EAG鉀離子通道在神經細胞的生理角色,我們利用酵母菌雙雜交系統,尋找與EAG鉀離子通道有相互作用的蛋白質。藉著檢視EAG鉀離子通道和其作用蛋白間的相互關係,以更進一步闡釋這些蛋白質交互作用的生理和病理意義。 離子通道功能的異常,已知和許多遺傳性疾病與癌症的產生有關,但是對於背後的致病機制仍有許多不清楚的地方。我們實驗室結合了細胞生物學、生物化學、和分子生物學等技術,探討離子通道蛋白質的構造與功能的關係,剖析鉀離子通道的生合成(Biosynthesis)與膜運送(Membrane trafficking)機制,研究基因突變之神經生理效應等,以進一步探究離子通道功能異常的致病機制,期能更了解電位控制鉀離子通道的生理與病理角色。 此外,我們實驗室利用培養的神經細胞,以分生和細胞生物學的方法,結合免疫螢光染色和共軛焦顯微鏡的觀察,研究神經細胞中分子的分布和變化。我們也利用培養的神經細胞,研究神經細胞受到傷害時的保護機制。
